Alzheimer's plaque components fight inflammation

In mice, bits of proteins can treat condition resembling multiple sclerosis

Tiny components of amyloid plaques, the notorious protein clumps found littering the brains of people with Alzheimer?s disease, might fight inflammation. Researchers report that several of these sticky protein fragments, or peptides, glom onto inflammatory compounds and reverse paralysis in mice that have a condition similar to multiple sclerosis. A fragment of tau protein, which shows up in other brain deposits in Alzheimer?s patients, has a similar effect.

When tested on blood taken from three MS patients, the tau peptide weeded out some inflammatory culprits there, too, researchers report in the April 3 Science Translational Medicine.

?This is a seriously good study. It opens up more questions than it answers,? says Jian-Guo Geng, a cell biologist at the University of Michigan in Ann Arbor who wasn?t part of the research team. ?But I don?t think we?re anywhere close to using these peptides for treatments.?

Amyloid is a broad term for clusters of protein in the brain, including those arising with the aid of misfolded versions of tau or another protein implicated in brain disease called a prion. Viewing amyloid-forming peptides as good guys runs against the scientific thinking, since amyloid plaques are a hallmark of Alzheimer?s disease. But study coauthor Lawrence Steinman, a neurologist at Stanford University, points out that the actual role of amyloid plaques in the disease is unclear. He suggests the tiny peptides holding the plaques together might have an alternative, useful role in the body.

Last year, Steinman and his team showed that injecting mice with amyloid improves symptoms in animals with the MS-like condition (SN: 9/22/2012, p. 14). Separately, Geng reported in 2012 that mice genetically engineered to make extra amyloid could fend off the MS-like disease, and that knocking out the amyloid-making capacity worsened symptoms in animals.

The new study goes a step further toward working out how amyloid fights disease in mice. In MS, inflammation is the enemy, damaging the fatty coatings on nerve sheaths in the central nervous system. A breakdown of that nerve insulation results in disordered nerve signaling and symptoms that can include weakness, loss of motor control, vision problems, loss of sensation and other problems.

Steinman and his colleagues identified several peptides, each only six amino acids in length, that seem to knock down inflammation when they twist to form sticky structures called fibrils. Steinman likens the peptide fibrils to sponges, because of their ability to bind to and contain compounds that trigger inflammation.

The researchers injected various peptides that can form amyloid in mice that had become disabled by the MS-like condition. Several of the peptides ? including fragments of heat shock proteins, tau protein and prion protein ? reversed the animals? paralysis during treatment. The effect later faded.

Blood samples from treated mice showed that heat shock and tau peptide fibrils had reduced levels of several proteins that cause inflammation, including interleukins 2 and 6. Compared with animals that received no fibril treatment, the animals? brain tissue had less inflammation.

When the scientists cultured human blood from MS patients with the tau fibril, the sticky peptide bound to dozens of compounds, including many molecules involved with inflammation.

The misfolded versions of tau, prions and amyloid protein can all form deposits in the brain, and it is the stickiness of fibrils that enables sheets of proteins to aggregate into these deposits, Steinman says. Those deposits have earned fibrils a nasty reputation.

?We have to keep an open mind,? he says. ?Most molecules are nuanced in function. In certain circumstances, they can cause harm or benefit.?

We've been thinking a lot lately about the chefs, musicians, comedians, actors and others whose career frequently takes them out on the road, into more airplanes, restaurants and hotel bars than they can count. We should have suspected that most of our Homecoming Meals participants would extoll the virtues of simple comfort food, but the consistency with which they've answered this way has delighted us. From Fabio's risotto to Jeni's unconventional ratatouille, simplicity totally rules. Today's edition is no exception: we caught up with "Top Chef" host, cookbook author and well-exposed food nerd, Padma Lakshmi.

Honestly it?s a simple bowl of lentils and rice. Or beans and rice. I?m very simple! Honestly most of my diet when I?m not working on ?Top Chef? is very simple, very plant based. So if it?s cold it will be a noodle soup and if it?s not cold it would probably be like beans and rice or lentils and rice. That?s really what I like.

As for flavors, I always think even dessert should have a hint of salt in it. But in the case of the lentils and rice, it?s actually homey, comfort, savory porridge called Kitchedi. And I add cumin to that and green chilies for a little heat. I also add sometimes some chopped dried plums for a hint of tartness and sweetness. I think ?- not to plug my own book but -? the title of my own book actually sums up what I think about food, I like everything a little tangy, a little tart, a little hot and a little sweet. [Padma?s book is called Tangy, Tart, Hot and Sweet: A World of Recipes for Everyday.] I think that?s the perfect bite in your mouth because it hits all the different parts of your palate.

Hybrid Memory Cube receives its finished spec, promises up to 320GB per second

The Hybrid Memory Cube Consortium has been almost too patient in developing a standard for for its eponymous technology -- efforts began 17 months ago -- but it at last has more than good intentions to show for its work. Its just-published HMC Specification 1.0 lets companies build platforms and RAM with 2GB, 4GB and 8GB chips incorporating the stacked, power-efficient technology, all without compatibility jitters from other supporters. The completed spec is a scorcher when living up to its full potential, too. With eight links, a memory cube can reach a peak 320GB/s (yes, that's gigabytes) of aggregate bandwidth -- more than a hair faster than the 11GB/s we often get from existing DDR3 memory.

The Consortium is teasing us with more. Although we'll have to wait until the second half of the year before HMC 1.0 products appear in earnest, the Consortium already has a next-gen blueprint due in early 2014 that should nearly double individual data link speeds (from 15Gbps to 28Gbps). While we'd like to see the group walk the walk with real products before it talks more talk, there's still a chance that some memory performance bottlenecks could vanish for a good, long while.

Lebanon has maintained a policy of "dissociation" from Syria's two-year-old conflict. But many Lebanese officials feel their country is increasingly at risk of being dragged into the civil war, which the United Nations says has killed 70,000 people.

BOSTON (AP) ? The Boston Bruins have acquired Jaromir Jagr from the Dallas Stars in an attempt to improve their shaky offense.

The 10th leading goal scorer in NHL history, who leads the Stars in points this season, Jagr was traded Tuesday for two prospects, forwards Lane MacDermid and Cody Payne, and a conditional second-round pick in this year's draft, the Stars announced.

The deal came five days after the Bruins thought they obtained forward Jarome Iginla from the Calgary Flames, only to learn Iginla had been traded to the Pittsburgh Penguins.

In his first season with the Stars, the 41-year-old Jagr has 14 goals and 26 points. Brad Marchand leads the Bruins with 14 goals and Patrice Bergeron is first with 31 points. Jagr has 679 career goals.

Euro outlook cut again after botched Cyprus bailout: Reuters poll

LONDON (Reuters) - Evaporating hopes for an economic recovery and last month's botched bailout of Cyprus persuaded analysts to chop their outlook again for the euro over the coming months, a Reuters poll showed.

Although the median expectation of more than 60 analysts surveyed over the last few days shows the euro trading around its current levels near $1.28 over the next few months, further out, the poll shows the currency weakening to around $1.25.

Overall, forecasts for the euro were about 2 cents weaker compared with last month's poll, which also reflected its roughly 6 percent depreciation against the dollar since the start of February.

The poor state of the euro zone economy was the chief reason why the euro is expected to weaken further, with surveys on Tuesday showing a worsening manufacturing slump in the currency bloc.

The median outlook for the euro was the weakest since September last year, when European financial markets were in turmoil as speculation grew that Spain would soon need a banking sector bailout, which it eventually got in December.

"We expect euro zone fundamentals to deteriorate further. This, combined with outflow pressures, should keep the euro's downward trend intact," said Camilla Sutton, chief currency strategist at Scotiabank in Toronto.

Asked which currencies investors were most likely to buy at the expense of the euro following the Cyprus bailout, 22 out of the 26 who answered said the U.S. dollar, with seven responses each for sterling and the Australian dollar.

"If the euro comes under further stress, we expect the growth-linked currencies like the Australian and Canadian dollars to benefit instead," said Emmanuel Ng, currency strategist at OCBC Bank in Singapore.

EURO BEARS OUT IN FORCE

Cyprus struck a 10-billion euro bailout deal with the European Union and International Monetary Fund last week, designed to untwine it from a failed banking sector that has long dominated its economy.

While tiny in economic terms, the chaotic bailout deal highlighted the political discord that has left Europe's economy rudderless, with the latest data showing signs of further deterioration rather than recovery.

The euro's recent decline versus the dollar reflects these poor economic fundamentals far more so than ebullient European stock markets. The FTSEurofirst 300 index has rallied more than 6 percent since the start of the year.

The poll showed the euro declining to $1.27 in six months, and $1.25 a year from now.

Bearishness about the euro's prospects among respondents was palpable, matching data from the Commodity Futures Trading Commission that showed traders increased their short bets against the euro through March.

Out of 58 contributors who took part in both the April and March foreign exchange polls, slightly more than half, 31, cut their 12-month outlook for the euro.

Fewer than a third of analysts expect the euro to be stronger in 12 months than its current level near $1.28.

Against sterling, analysts expect the euro to trade near 85 pence over the next year.
