United States "not threatened" by China's surge in Africa -Obama

PRETORIA (Reuters) - The United States does not feel threatened by the growth of trade and investment in Africa by China and other emerging powers, U.S. President Barack Obama said on Saturday.

"I don't feel threatened by it. I feel it's a good thing," Obama told a news conference during a visit to South Africa.

He said the more countries invested in Africa the more the world's least developed continent could be integrated into the global economy.

"I want everybody playing in Africa. The more the merrier," Obama said.

Obama is embarked on an eight-day trip through Africa to promote trade and business ties between the United States and the continent.

However, he has had to defend his administration against charges it has lagged its predecessors in the level of engagement with Africa, leaving countries like China to reap the economic benefits of a more aggressive approach.

The president's aides have argued that Obama has had two wars and a deep economic crisis to deal with since he took office in 2009.

Obama has also said that U.S. interactions with Africa have included goals of social and political development, unlike those of China, which he said were more narrowly focused on commercial benefits.

"A lot of people are pleased that China is involved in Africa," he told reporters travelling with him on Friday.

"On the other hand, they recognise that China's primary interest is being able to obtain access for natural resources in Africa to feed the manufacturers in export-driven policies of the Chinese economy."

That relationship makes Africa an exporter of raw materials but does not create jobs in Africa and is not a sustainable model over the long-term, he added.

Berlin police shoot dead naked man wielding knife

BERLIN (AP) ? Police have fatally shot a naked man who brandished a knife at officers in a landmark Berlin fountain.

Police spokesman Stefan Redlich said passers-by reported early Friday that a man was behaving strangely and carrying a long knife in the Neptune fountain, near Berlin's city hall.

Officers tried to persuade the man to put the knife away, but instead he started cutting himself. A policeman climbed into the water to try to stop him, whereupon the man advanced on the officer with the knife.

Another officer shot the man after he ignored calls to back off. He died at the scene.

Police say an autopsy was being conducted to determine whether the man, apparently aged around 20, died from the shot or his self-inflicted injuries.

It seems like the recent stock market tumble, and the perception that interest rates are rising, are, at most, background noise to American car buyers.

Sales of new cars and trucks continue to move along at a steady clip during June, according to new estimates from J.D. Power & Associates and LMC Automotive. A monthly sales forecast based on direct dealer data indicates new-vehicle retail sales are showing no signs of letting up at the start of the summer selling season.

New-vehicle retail sales in June are projected to come in at 1,118,800 vehicles, which represent a Seasonally Adjusted Annualized Rate of 13.2 million units, a healthy increase of 500,000 from the May SAAR. Retail transactions are the most accurate measure of true underlying consumer demand for new vehicles.

Total light-vehicle sales in June 2013 are expected to grow by 12 percent from June 2012 to 1,380,800 units. Fleet sales in June are just 19 percent of total sales. Fleet volume for the month is projected at 262,000 units.

Adding together retail and fleet business and the overall SAAR is expected to reach 15.7 million units this month. That?s a big jump from the 14.5 million vehicles sold in 2012 ? a five-year high ? and nudges by even the most optimistic forecasts for 2013, which general had set a high of around 15.5 million sales this year.

The strong selling pace continues to be matched by strong transaction prices. Thus far in June, the average transaction price of new vehicles ? what customers actually spend when both incentives and options are included ?is $28,900, the highest figure ever for June.

While sales overall are strong, not all segments are selling at the same pace. Sales of premium vehicles account for just 11.7 percent of new-vehicle retail sales thus far in June, down from 12.9 percent in June 2012.

?Although the premium segment growth has lagged non-premium, there is some good news for the industry in that the average price of premium vehicles in June is $47,000, up almost 4 percent from June 2012,? said John Humphrey, J.D. Power senior vice president of the global automotive practice. ?New premium vehicles entering the market late this year will also help bolster sales through the second quarter of 2014.?

Among other new models due for launch are the all-new Mercedes-Benz S-Class and a trio of luxury diesels from Audi.

The underperformance of premium light-vehicle sales is largely due to the age of the models in these segments. J.D. Power calculates that the average age ? the number of months the vehicle has been in the market since it was introduced or redesigned ? of premium models sold in the second quarter of 2013 was 43 months. In comparison, the average age of non-premium models, excluding pickup trucks is only 34.5 months.

Hyundai America chief executive officer John Krafcik noted last week that competition in the auto industry is very fierce and forces manufacturers to intensify efforts to win over customers. Even the threat of higher interest rates hasn?t undermined the market?s momentum, he said.

Competition in the mid-sized segment has been particularly fierce in recent months with Toyota, Honda, Nissan, Ford, General Motors, Hyundai, Volkswagen and Kia all introducing new or substantially updated models. The compact and subcompact segment also have seen a flood of new entries.

J.D. Power expects that by the second quarter of 2014, the average age of premium products will fall to just 33 months, as new and redesigned products enter the marketplace.

LMC Automotive continues to hold the outlook for total light-vehicle sales in 2013 at 15.4 million units, but has increased its forecast for retail light-vehicle sales to 12.6 million units from 12.5 million units, as retail sales growth expands.

Consumer spending rebounds, jobless claims fall

WASHINGTON (Reuters) - Consumer spending rebounded in May and new applications for unemployment benefits fell last week, suggesting the economy remained on a moderate growth path.

The Commerce Department said on Thursday consumer spending increased 0.3 percent last month after a revised 0.3 percent drop in April. Consumer spending in April was previously reported to have declined 0.2 percent.

Last month's spending increase was in line with economists' expectations. When adjusted for inflation, consumer spending rose 0.2 percent last month after dipping 0.1 percent in April.

In a separate report, the Labor Department said initial claims for unemployment benefits fell 9,000 to a seasonally adjusted 346,000. The four-week moving average for new claims, which irons out week-to-week volatility, fell 2,750 to 345,750.

U.S. stock index futures slightly added to earlier gains after the data. U.S. Treasuries prices extended price gains and yields fell to session lows, while the dollar pared gains against the yen.

Recent data, including housing, regional factory activity, business spending plans and consumer confidence, have pointed to an economy that is regaining some speed after stumbling early in the second quarter.

That is broadly supportive of the view the Federal Reserve expressed last week that the downside risks to the economy's outlook have waned. Fed Chairman Ben Bernanke said the U.S. central bank could start scaling back on the pace of its monthly bond purchases this year.

Consumer spending accounts for 70 percent of U.S. economic activity. Though the pace of spending has slowed from the 2.6 percent annual rate notched in the first three months of the year, consumers will likely continue to drive growth in the second quarter.

The firming growth theme held as other details of the Commerce Department report showed income grew 0.5 percent last month, the largest gain since February, after nudging up 0.1 percent in April. That reflects a steady pace of job gains.

Households also saved a bit more last month, lifting the saving rate to a five-month high of 3.2 percent.

There was also a bit of inflation in the economy last month, pointing to some pick-up in demand.

A price index for consumer spending inched up 0.1 percent in May after declining two straight months. A core reading that strips out food and energy costs also rose 0.1 percent after being flat in April.

Over the past 12 months, inflation rose 1 percent, still below the Fed's 2 percent target. The index had increased only 0.7 percent in the period through April.

Core prices were up 1.1 percent from a year ago after rising by the same margin in April.

Falling healthcare costs are dampening inflation pressures. Bernanke has said the weak healthcare costs were likely to transitory.

Gene deletion affects early language and brain white matter

June 27, 2013 ? A chromosomal deletion is associated with changes in the brain's white matter and delayed language acquisition in youngsters from Southeast Asia or with ancestral connections to the region, said an international consortium led by researchers at Baylor College of Medicine. However, many such children who can be described as late-talkers may overcome early speech and language difficulties as they grow.

The finding involved both cutting edge technology and two physicians with an eye for unusual clinical findings. Dr. Seema R. Lalani, a physician-scientist at BCM and Dr. Jill V. Hunter, professor of radiology at BCM and Texas Children's Hospital, worked together to identify this genetic change responsible for expressive language delay and brain changes in children, predominantly from Southeast Asia.

Lalani, assistant professor of molecular and human genetics at BCM, is a clinical geneticist and also signs out diagnostic studies called chromosomal microarray analysis, a gene chip that helps identify abnormalities in specific genes and chromosomes, as part of her work at BCM's Medical Genetics Laboratory.

"I got intrigued when I kept seeing this small (genomic) change in children from a large sample of more than 15,000 children referred for chromosomal microarray analysis at Baylor College of Medicine. These children were predominantly Burmese refugees or of Vietnamese ancestry living in the United States. It started with two children whom I evaluated at Texas Children's Hospital and soon realized that there was a pattern of early language delay and brain imaging abnormalities in these individuals carrying this deletion from this part of the world. Within a period of two to three years, we found 13 more families with similar problems, having the same genetic change. There were some children who obviously were more affected than the others and had cognitive and neurological problems, but many of them were identified as late-talkers who had better non-verbal skills compared to verbal performance," said Lalani. Hunter, helped in determining the specific pattern of white matter abnormalities in the MRI (magnetic resonance imaging) scans in children and their parents carrying this deletion. Most of the children either came from Southeast Asia or were the offspring of people from that area. (White matter is the paler material in the brain that consists of nerve fibers covered with myelin sheaths.)

Now, in a report that appears online in the American Journal of Human Genetics, Lalani, Hunter and an international group of collaborators identify a genomic deletion on chromosome 2 that is associated with bright white spots that show up in an MRI in the white matter of the brain . The chromosomal deletion removes a portion of a gene known as TM4SF20 that encodes a protein that spans the cellular membrane. They do not know yet what the function of the protein is. They found this genetic change in children from 15 unrelated families mainly from Southeast Asia.

"This deletion could be responsible for early childhood language delay in a large number of children from this part of the world," says Lalani.

She credits Dr. Wojciech Wiszniewski, an assistant professor of molecular and human genetics at BCM with doing much of the work. Wiszniewski has an interest in genomic disorders and is working under the mentorship of Dr. James R. Lupski, vice chair of the department of molecular and human genetics.

Lupski said, "Professor Lalani has made a stunning discovery in that she provides evidence that population-specific intragenic CNV (copy number variation -- a deletion or duplication of the chromosome) can contribute to genetic susceptibility of even common complex disease such as speech delay in children."

"In a way, this is a good news story," said Hunter. There is evidence from family studies that some of these children may do quite well in the future, said Lalani.

Lalani elaborates. "This is a genetic change that is present in 2 percent of Vietnamese Kinh population (an ethnic group that makes up 90 percent of the population in that country)," she said. "In the 15 families we have identified, all children have early language delay. Some are diagnosed with autism spectrum disorder, and if you do a brain MRI study, you find white matter changes in about 70 percent of them. We have found this change in children who are Vietnamese, Burmese, Thai, Indonesian, Filipino and and Micronesian. It is very likely that children from other Southeast Asian countries within this geographical distribution also carry this genetic change."

Because these are all within a geographic location, she suspects that there is an ancient founder effect, meaning that at some point in the distant past, the gene deletion occurred spontaneously in an individual, who then passed it on to his or her children and to succeeding generations.

"It is important to follow these children longitudinally to see how these late-talkers develop as they grow," said Lalani. "We have also seen this deletion in children whose parents clearly were late-talkers themselves, but overcame the earlier problems to become doctors and professionals. The variability within the deletion carriers is fascinating and brings into question genetic and environmental modifiers that contribute to the extent of disease in these children.

Language delays mean that they may speak only two or three words at age 2, in comparison to other children who would generally have between 75-100 word vocabulary by this age. While there is evidence that children with this deletion may catch up, it is unclear if they continue to have better non-verbal skills than verbal skills. It is also unclear how the specific brain changes correlate with communication disorders in these children.

In fact, when doctors check the parents of these children, they often find similar white matter changes in the parent carrying the deletion. "Young parents in their 30s should not have age-related white matter changes in the brain and these changes should definitely not be present in healthy children," said Lalani. Hunter said they are not sure how the gene variation relates to the changes in brain white matter and how all of these result in delay in language.

Emmys: MTV 'Catfish's' Nev Schulman Says Manti Te'o Proved His Reality Show's Legit

LOS ANGELES (TheWrap.com) - With a second season of MTV's "Catfish: The TV Show" debuting Tuesday at 10/9c, Nev Schulman has become one of reality television's most interesting people. He has seen himself catapulted from indie documentary filmmaker to TV's most well-known internet scam buster/matchmaker.

His reality career was created and then boosted by serendipitous story turns, beginning with the success of "Catfish," the 2010 doc in which Schulman's brother Ariel and friend Henry Joost filmed his online romance with a woman who turned out to have created a fake identity.

The MTV reality series of the same name, in which Shulman and Max Joseph travel the country arranging in-person meetings between folks engaged in online-only romances, debuted last November to some buzz.

It had a further spotlight shone on it in January, when college football player Manti Te'o's online romance was revealed to be a sham. The show's ratings rose 30 percent with the news, and Nev found himself in great demand by reporters seeking his explanation for how such a thing could happen.

Had you ever planned to have a career in reality TV? I never thought this would be the career path that I would end up taking. The movie came as a big surprise, and its success was also very unexpected.

The TV show sort of hatched organically from that. And now that I'm part of the entertainment industry, I feel a lot more opportunity to continue doing that, but it was never anything that I pursued initially.

Some viewers wonder if your stories are doctored, since reality shows already carry with them the suspicion that they're scripted. How do you deal with that? For those people who don't think the show's legit, I need only to point to Manti Te'o as a case study to prove that this is a very real thing. It's happening frequently, and basically if you turn your cameras on and go out in the world looking for it, it's there. We didn't have to script it; we didn't have to make anything up. It found us.

When the Te'o story broke, you began to investigate his case. What was it like to compete with the press at the time? I had an interesting leg up, in a small way, because I had received correspondence from some of the people who had been involved in the story. Months earlier, they had reached out to me, prior to any of the news happening. Unfortunately, I hadn't seen those emails because they had been buried under many other emails similar in nature. But when the story broke, I reached back out to those people, and I definitely had an interesting inside angle.

But in the end, I didn't have much of an advantage over the press except that a tremendous amount of people reached out to me when they heard about the story because they thought they were in communication with or "being catfished" by the same person. That's when everybody started to realize this was not some small, strange, once-in-a-while kind of thing.

Do you plan on expanding into other reality series?

The exciting thing right now is that I'm speaking at a lot of schools, colleges and universities and not just about the show and my experiences in the film. Generally, more about the best practices in being online and protecting yourself and being accountable and honest. Sort of the unofficial rules of the internet that no one seems to have been able to establish yet.

But yeah, I'm definitely interested in working on more shows as a host and also as a producer. I think it's a field I'm well suited for.

Army to cut brigades at 10 US bases

WASHINGTON (AP) ? The Army will eliminate at least 12 combat brigades, relocate thousands of soldiers and cancel $400 million in construction projects as the first wave of federal budget cuts takes aim at military communities around the country.

In a massive restructuring, Army leaders said Tuesday that they will slash the number of active duty combat brigades from 45 to 33, as the service moves forward with a longtime plan to cut the size of the service by 80,000. And they warned that more cuts ? of as many as 100,000 more active duty, National Guard and Reserve soldiers ? could be coming if Congress allows billions of dollars in automatic budget cuts to continue next year.

The sweeping changes would eliminate brigades ? which number from 3,500 to 5,000 troops ? at 10 Army bases in the U.S. by 2017, including those in Texas, Kentucky, Georgia, Colorado, North Carolina, New York, Kansas and Washington. The Army will also cut thousands of other jobs across the service, including soldiers in units that support the brigades, and two brigades in Germany have already been scheduled for elimination.

Gen. Ray Odierno, Army chief of staff, said one additional brigade will likely be cut, but no final decisions have been made.

"I know in the local communities it will have its impact," Odierno told reporters Tuesday. "But we've done our best to reach out to them so they understand what the impacts are. We've tried to make it as small an impact as possible for as many communities as we could."

Members of Congress, meanwhile, expressed concerns about the prospects for greater cuts down the road.

Rep. Howard P. "Buck" McKeon, R-Calif., chairman of the House Armed Services Committee, said his panel "will carefully examine the implications of this initial restructuring, but we all must understand that this is only the tip of the iceberg, much deeper cuts are still to come."

The Army is being reduced in size from a high of about 570,000 during the peak of the Iraq war to 490,000 as part of efforts to cut the budget and reflect the country's military needs as the wars in Iraq and Afghanistan end. Odierno said that the potential 100,000 more would be spread out across the active duty, Guard and Reserves, and that there also could be reductions in the Army's 13 aviation brigades.

While the personnel cuts may have less impact at some of the Army's larger bases such as Fort Hood in Texas and Fort Bragg in North Carolina, they could be more painful for communities around some of the smaller installations such as Fort Knox, where currently only one brigade is based.

The other seven U.S. bases that will lose a brigade are: Fort Bliss in Texas, Fort Campbell in Kentucky, Fort Carson in Colorado, Fort Drum in New York, Fort Riley in Kansas, Fort Stewart in Georgia, and Joint Base Lewis-McChord in Washington. Soldiers in the deactivated brigades would be transferred to other units.

Odierno said the Army tried to spread out the cuts geographically. He said Fort Knox scored the lowest in military value, but insisted the reduction was not the first step toward closing the base. He noted that about 4,000 civilians workers had been added there, as well as the Army's recruiting command.

The overall cut in size has been known for more than a year, and Army leaders have been working on how to manage the reduction, conducting local community meetings across the country and releasing an extensive study on the issue earlier this year.

Under the plan announced Tuesday, the Army will increase the size of its infantry and armor brigades by adding another battalion, which is between 600-800 soldiers. Adding the battalion was a recommendation from commanders in Iraq and Afghanistan who said it would beef up the fighting capabilities of the brigades when they go to war.

Odierno said he continues to hope that he will be able to cut the 80,000 soldiers largely through voluntary departures. He said he believes he will have to force several hundred officers to leave in order to get the proper number of soldiers at various ranks. But, if the automatic cuts go forward, Odierno said he would likely have to force soldiers out of the Army.

These initial brigade cuts do not affect National Guard or Reserve units.

Officials said the decisions on the cuts were based on a variety of factors including required training resources, ranges, air space and infrastructure, as well as the need to put units near leadership and headquarters units.

Heavyweights Matt Mitrione, Brendan Schaub exchange nasty insults on Twitter

UFC heavyweights Matt Mitrione and Brendan Schaub were once teammates on "The Ultimate Fighter." Since they both have backgrounds in football, that common ground would make for a good friendship, right? Well, not so much. They are fighting at UFC on Fox 8, and they exchanged some nasty insults on Twitter on Monday night.

Here's a sampling of what they had to say each other:

It got uglier, as they used language we're not fond of around here, as well as insults involving Mitrione's wife and comparing Schaub's looks to an actor with Down's Syndrome. The discipline Schaub is speaking of is Mitrione's short suspension for his comments about trans fighter Fallon Fox.

Israeli aircraft strike targets in Gaza Strip

JERUSALEM (AP) ? Israeli aircraft pounded targets in the Gaza Strip early Monday after rockets were fired at Israel from the territory, the military said, unsettling a tenuous cease-fire between Israel and Hamas.

The military said its aircraft struck two weapons storage facilities and a rocket launch site. No injuries were reported.

Rocket fire from Gaza has declined since Israel carried out an eight-day military campaign last November in response to frequent attacks. An Egyptian-brokered cease-fire has largely held, but sporadic fire still persists.

No militant group claimed responsibility for the rocket launch, but Israel said it holds Hamas, which rules the coastal territory, accountable.

"Last night's rocket attack is an intolerable act of aggression against Israel and its civilians. Hamas is held accountable for all acts of terrorism deriving from the Gaza Strip," said military spokesman Lt. Col. Peter Lerner.

Israeli police spokesman Micky Rosenfeld said a total of six rockets were fired at Israel overnight, causing no damage or injuries. The military said two rockets were intercepted by the "Iron Dome" missile defense system. The other four landed in open areas.

Israeli Prime Minister Benjamin Netanyahu responded to the rocket fire, saying "my policy is to harm whoever tries to harm us ... That is how we will work and will continue to act against threats that are close and threats that are far."

In Gaza, Hamas Prime Minister Ismail Haniyeh said he would not be intimidated by Israel's strikes. "Any Israeli aggression does not scare the Palestinian people," he said.

Israel last struck in Gaza in April, when its aircraft hit and killed a top militant in a shadowy al-Qaida-influenced group who had been involved in a rocket attack. The rocket was launched from Egypt's Sinai Peninsula, where Gaza militants are believed to operate. A rocket last landed in Israel in May.

Meanwhile, Israeli police said that vandals slashed the tires of 21 cars in an Arab neighborhood of east Jerusalem. The vandals also scribbled slogans on nearby walls.

It was the latest in a wave of crimes linked to Jewish extremists that has targeted mosques, churches, monasteries, dovish Israeli groups and even Israeli military bases to protest what they perceive as the Israeli government's pro-Palestinian policies in the West Bank. Vandals call the attacks the "price tag" for the policies they oppose. Last week vandals struck an Arab village outside of Jerusalem that has been a model of coexistence in Israel.

Rosenfeld said police were investigating. He said no arrests have been made in the recent string of similar crimes.

Diet may affect Alzheimer's disease risk

June 17, 2013 ? The lipidation states (or modifications) in certain proteins in the brain that are related to the development of Alzheimer disease appear to differ depending on genotype and cognitive diseases, and levels of these protein and peptides appear to be influenced by diet, according to a report published Online First by JAMA Neurology.

Sporadic Alzheimer disease (AD) is caused in part by the accumulation of ?-amyloid (??) peptides in the brain. These peptides can be bound to lipids or lipid carrier proteins, such as apolipoprotein E (ApoE), or be free in solution (lipid-depleted [LD] ??). Levels of LD ?? are higher in the plasma of adults with AD, but less is known about these peptides in the cerebrospinal fluid (CSF), the authors write in the study background.

Angela J. Hanson, M.D., Veterans Affairs Puget Sound Health Care System and the University of Washington, Seattle, and colleagues studied 20 older adults with normal cognition (average age 69 years) and 27 older adults with amnestic mild cognitive impairment (average age 67 years).

The patients were randomized to a diet high in saturated fat content (45 percent energy from fat, greater than 25 percent saturated fat) with a high glycemic index or a diet low in saturated fat content (25 percent of energy from fat, less than 7 percent saturated fat) with a low glycemic index. The main outcomes the researchers measured were lipid depleted (LD) ??42 and ??40 and ApoE in cerebrospinal fluid.

Study results indicate that baseline levels of LD ?? were greater for adults with mild cognitive impairment compared with adults with normal cognition. The authors also note that these findings were more apparent in adults with mild cognitive impairment and the ?4 allele (a risk factor for AD), who had higher LD apolipoprotein E levels irrespective of cognitive diagnosis. Study results indicate that the diet low in saturated fat tended to decrease LD ?? levels, whereas the diet high in saturated fat increased these fractions.

The authors note the data from their small pilot study need to be replicated in a larger sample before any firm conclusions can be drawn.

"Overall, these results suggest that the lipidation states of apolipoproteins and amyloid peptides might play a role in AD pathological processes and are influenced by APOE genotype and diet," the study concludes.

Editorial: Food for Thought

In an editorial, Deborah Blacker, M.D., Sc.D., of the Massachusetts General Hospital/Harvard Medical School, Boston, writes: "The article by Hanson and colleagues makes a serious effort to understand whether dietary factors can affect the biology of Alzheimer disease (AD)."

"Hanson et al argue that the changes observed after their two dietary interventions may underlie some of the epidemiologic findings regarding diabetes and other cardiovascular risk factors and risk for AD. The specifics of their model may not capture the real underlying biological effect of these diets, and it is unclear whether the observed changes in the intermediate outcomes would lead to beneficial changes in oligomers or plaque burden, much less to decreased brain atrophy or improved cognition," she continues.

"At some level, however, the details of the biological model are not critical; the important lesson from the study is that dietary intervention can change brain amyloid chemistry in largely consistent and apparently meaningful ways -- in a short period of time. Does this change clinical practice for those advising patients who want to avoid dementia? Probably not, but it adds another small piece to the growing evidence that taking good care of your heart is probably good for your brain too," Blacker concludes.

New compound excels at killing persistent and drug-resistant tuberculosis

June 17, 2013 ? An international team led by scientists at The Scripps Research Institute (TSRI), the Howard Hughes Medical Institute and Albert Einstein College of Medicine of Yeshiva University has identified a highly promising new anti-tuberculosis compound that attacks the tuberculosis (TB) bacterium in two different ways.

"These findings represent an effort to help solve one of the major global health crises of our time -- the resurgence of TB and its dangerous drug-resistant strains," said Peter G. Schultz, the Scripps Family Chair Professor of Chemistry at TSRI, who was senior author of the study with William R. Jacobs, Jr., member of the Howard Hughes Medical Institute and professor of microbiology & immunology and of genetics at Albert Einstein College of Medicine.

"In cell cultures and in mice, this compound showed powerful activity against ordinary active TB bacteria, non-replicating TB bacteria and even extensively drug-resistant TB strains," said Feng Wang, a member of the Schultz lab at TSRI and first author of the study with Dhinakaran Sambandan of the Jacobs lab and Rajkumar Halder of the Schultz lab.

The paper appears in this week online ahead of print in an Early Edition of the Proceedings of the National Academy of Sciences.

Global Health Crisis

Although isoniazid and rifampin, the two front-line TB drugs, came into use in 1952 and 1967 respectively, new TB infections still occur at the rate of roughly one per second. At any moment about a third of the existing human population is infected -- mostly with inactive, latent TB, although active TB still kills over one million people each year. Russia, Africa, China and Southeast Asia have been especially hard hit by the epidemic.

Increased urbanization, public health complacency and immunity-weakening HIV have been major enablers of TB's spread in recent decades. But the bacterium that causes TB -- Mycobacterium tuberculosis (Mtb) -- also happens to be unusually well adapted for persisting in humans. Among other strategies, it frequently reverts to a dormant, non-replicating state and also creates attack-resistant cell colonies called biofilms, which contain a high proportion of non-replicating TB.

Compared to ordinary, fast-replicating TB, these other forms of TB are much less susceptible to existing drugs. Effective TB therapy thus requires months to years of regular dosing. But many patients quit before completing such long courses of treatment and end up incubating drug-resistant TB strains. Some strains are now "extensively drug-resistant" (XDR) and virtually untreatable -- and usually fatal.

Killing the Persisters

"The big challenge here has been to find a drug that clears TB infection more quickly, which means it has to be effective against both replicating and non-replicating TB," said Wang, now also a scientist at the California Institute for Biomedical Research (CALIBR), a non-profit organization founded by Schultz for the early-stage development of new medicines.

Most existing TB drugs work poorly against non-replicating TB, having been developed principally for their ability to kill actively replicating TB. Wang therefore set up a different kind of screening test -- one to detect compounds that block TB's persistence-related ability to form biofilms.

Because experiments with live TB require a special (level 3) biosafety facility, Wang used a related but non-disease-causing mycobacterium for his initial, high-throughput test. Screening a diverse library of 70,000 compounds, he quickly found one, dubbed TCA1, that stood out for its ability to inhibit mycobacterial biofilms.

Tests in Jacobs's biosafety level 3-certified laboratory confirmed that TCA1 also has powerful activity against TB. "Surprisingly, it turned out to kill both non-replicating and replicating TB," Wang said.

In cell culture tests, TCA1 on its own killed more than 99.9% of ordinary, actively replicating TB bacteria within three weeks, and in combination with isoniazid or rifampin, could kill 100% within that period. TCA1 also showed strong effectiveness against drug-resistant TB strains, removing all signs of one common strain within a week when combined with isoniazid. Against a highly fatal "super-bug" strain from South Africa, which resists all conventional TB drugs, the new compound on its own had a kill rate of more than 99.999% within three weeks.

As expected, TCA1 also showed potent effects against non-replicating TB. Tests in mice confirmed TCA1's effectiveness and suggested that the combination of TCA1 and isoniazid could be more powerful than existing drug regimens. TCA1 showed no sign of toxicity or adverse side effects in cell culture and mouse experiments, and also showed almost no tendency to induce drug resistance in TB.

A Complex Mechanism

Working with the laboratories of Gurdyal S. Besra and Klaus F?tterer at the University of Birmingham, UK, Katarina Mikusova at Comenius University in Slovakia, and Kai Johnson at Ecole Polytechnique F?d?rale de Lausanne (EPFL) in Switzerland, the team next did structural and biochemical analyses to determine how the new compound kills Mtb so efficiently.

The researchers found it works in an apparently unique way, largely by targeting two Mtb enzymes, one supporting TB replication and the other TB dormancy and persistence. "I don't know of any other antibiotic that kills replicating bacteria through one pathway and non replicating bacteria through another, as this one does," Wang said.

Now funded by the Global Alliance for TB Drug Development, Wang and his colleagues are working to devise improved variants of TCA1. "We already have analogs of TCA1 that are more potent and look very promising as TB drug candidates," said Wang.

Assuming that preclinical tests are completed successfully, he added, the group hopes to find a pharmaceutical company partner to sponsor clinical trials in TB patients.

The study was funded in part by the National Institutes of Health (grants AI26170 and A10-97548), the European Community's Seventh Framework Programme (260872), the Global Alliance for TB Drug Development and the Albert Einstein College of Medicine Center for AIDS Research (AI0-51519).

Trans-US solar plane reaches Dulles

A solar-powered plane has touched down in Washington DC, ending the penultimate leg of a first-of-its-kind bid to cross the US on solar power.

The Solar Impulse vehicle landed at the capital's Dulles airport at 00:15 local time (04:15 GMT).

The plane will remain in Washington until early July, when it will take off bound for New York - the final leg of the "Across America" project.

On Sunday, the plane will be on view to the public at Dulles Airport.

This is the location of the US National Air and Space Museum's Steven F Udvar-Hazy Center.

The fourth leg of the journey from St Louis was re-routed via Cincinnati at the last minute, as high winds and air traffic would have made the direct journey longer than the team's prescribed limit of 24 hours for one pilot to be at the controls.

After a 15-hour, 14-minute hop, the HB-SIA prototype craft landed at Cincinnati's Municipal Lunken airport on Friday evening in order to allow a change of pilots - Bertrand Piccard taking over from Andre Borschberg.

The segment from Cincinnati's Municipal Lunken airport to Dulles lasted 14 hours and four minutes.

On landing, Andre Borschberg said that "with the successful completion of these last four US flights, we have shown that we are capable of coping with challenging meteorological conditions for our weather-sensitive plane and for our ground operations, and that we could find each time the right solutions to move forward. It has been a succession of fruitful learnings preparing us for the 2015 world tour."

Piccard's and Borschberg's intention is to build a bigger plane than the HB-SIA prototype and fly it first across an ocean and then around the globe.

The HB-SIA craft already holds the world record for the longest manned solar-powered flight at 26 hours. The aeroplane's other records include the first international flight of a manned solar-powered plane in 2011, and first inter-continental flight in 2012.

The Across America project coincides with Piccard's and Borschberg's Clean Generation Initiative, an effort to encourage policy-makers and businesses to develop and adopt sustainable energy technologies.

OCD & Your Cat, Dog or Family Pet | Psych Central

OCD & Your Cat, Dog or PetMy son Dan suffered from obsessive-compulsive disorder so severe he could not even eat, and his anxiety levels were often so high, he could barely function. It would have been ludicrous for me to suggest he try yoga, or meditation, or any other stress reduction technique to help him feel better when, in fact, he could hardly get off the couch.

But he could pet our cats.

Our beautiful cats, Smokey and Ricky, both so lovable with distinct personalities, helped Dan immensely during those dark days. Whether they sat on his lap, curled up near him on the couch, or let him hold them, they allowed him to relax and brought him momentary peace. Sometimes they purred so loudly they sounded like engines revving, and this soothed Dan. Other times they would engage in various cat-like antics, inciting a rare, but oh-so-cherished laugh from our son.

They didn?t bombard him with questions, asking if he was okay, or if he was hungry, or what was wrong. They were just there with Dan, and for a short time, his focus was diverted from his obsessions and compulsions. Our pets were able to care for Dan in a way the rest of our family could not.

An article in the April 15, 2013 issue of Time magazine explored how animals grieve. I found it fascinating, and no matter how you might interpret the various studies discussed in the article, I think it is hard to argue with the belief that animals do indeed form relationships, and are empathetic. What more does it take to comfort someone?

For those obsessive-compulsive disorder (OCD) sufferers who struggle with germs and contamination issues, caring for a pet can elicit many triggers. Cleaning a litter box, letting a dog lick your face, or having to tend to a sick pet are just a few examples of what OCD sufferers might have to deal with. Surprisingly, I have heard from many with OCD who are amazed themselves that these situations do not cause their OCD to spring into action. Could it be that their love for their pets transcends the fear and anxiety of OCD?

When my son moved into his own apartment last year, one of the first things he did was foster a cat from a shelter. He has always been an animal lover, and was looking for a furry friend to keep him company. As he knows, life is full of surprises, and come to find out, his new companion has a host of medical problems and needs to take medication to control her seizures.

Instead of returning the cat to the animal shelter (something I very well might have done), he has embraced his role as her caretaker. Whether we have OCD or not, I believe this experience of putting another?s needs ahead of our own is worthwhile. Focusing outward instead of inward gives us a different perspective on our own lives and challenges.

So it works both ways. We take care of our beloved pets, and they take care of us. Whether our furry friend is a specially trained service dog who can sense an imminent anxiety attack (yes, it?s possible!) or an adored rabbit, pets can benefit us all in countless ways. They require us to slow down our lives, they make us laugh, and they give us unconditional love. And for those who are suffering, they provide the much-needed comfort and serenity that often can?t be found elsewhere.

On Saturday, we examined the many recent trades of aces that did not work out for the trading team, either because the return didn?t appear sufficient at the time or because the prospects acquired didn?t pan out (see story).

But several trades of aces have worked out for the team that unloaded its prized piece. It?s not as if each and every trade was a failure.

Let?s have a look:

July 27, 2012 ? Brewers trade Zack Greinke to Angels for Jean Segura, Johnny Hellweg and Ariel Pena

Segura was the 55th best prospect in baseball heading into last season, according to MLB.com. He wasn?t a sure thing, but in him the Brewers got their shortstop of the future who has power, speed and the ability to hit balls all over the field.

Hellweg, a 6-foot-9 right-handed pitcher, is a project who strikes a lot of batters out but walks way too many.

Pena is already 24 and has limited upside, but has pitched very well at Double A for Milwaukee this season.

Segura was the centerpiece of the deal and his presence makes it worthwhile for Milwaukee regardless of Hellweg and Pena working out. The Brewers got Segura for three months of Greinke.

The Phillies need to get a player like Segura ? not one with his skills necessarily, but his readiness for the majors and overall talent ? in any trade of Lee.

Meanwhile, Corbin is 9-0 with a 2.28 ERA this season for Arizona at age 23, and Skaggs was MLB.com?s 10th-rated prospect in all of baseball heading into the season.

Saunders gave the Diamondbacks 67 starts with an ERA under 4.00, so all around this was a great trade for Arizona, and one that inspires hope that solid returns can be had for aces. Of course, this one was built entirely around pitching. The Phillies need impact bats more than they need young arms.

Dec. 14, 2007 ? Athletics trade Dan Haren and Connor Robertson to Diamondbacks for Carlos Gonzalez, Brett Anderson, Chris Carter, Aaron Cunningham, Dana Eveland and Greg Smith

An enormous deal, and a rare case featuring quantity and quality. Gonzalez turned into an offensive monster, albeit not in Oakland. He did, however, bring Matt Holliday to the A?s, which in turn brought Oakland another set of prospects (who didn?t work out).

Anderson can?t stay healthy, but is a No. 2 starter when he is. Carter has traveled to various organizations and is finally getting a chance to play every day this season in Houston, where he?s on pace for 33 homers and 221 strikeouts.

The others ? Cunningham, Eveland and Smith ? have all been bit parts on major-league rosters. Better than not panning out at all.

On the whole, this was a great return for the A?s, and if they didn?t have to worry about finances and were able to keep CarGo, it would look much, much better.

Dec. 9, 2012 ? Rays trade James Shields, Wade Davis and Elliot Johnson to Royals for Wil Myers, Jake Odorizzi, Mike Montgomery and Patrick Leonard

The precedent for the Phillies acquiring a Jurickson Profar, Oscar Taveras or Xander Bogaerts.

Tampa Bay dealt Shields, a pitcher who is less than an ace but more than a No. 2, for the minor-league player of the year and a consensus top-five overall prospect in Myers. He will likely be in the middle of Tampa?s order with Evan Longoria for 5-7 years.

The Rays also landed righty starter Odorizzi, a 23-year-old former first-round pick who has had excellent strikeout and walk numbers in the minors. Montgomery was formerly a top prospect in Kansas City, but control issues ended that. He still has plenty of upside.

Shields is younger than Lee, but is a lesser pitcher who is under control for 2014 only. After that, the Royals will have to pony up the dough if they want to keep him, which they probably have to based on what they traded. Kind of takes leverage away.

In any event, Lee has way more trade value than Shields did, even with his age and price tag. As we stated last week, any team in baseball would jump at the chance to sign Lee for three years, $62.5 million if given the opportunity tomorrow.

The obvious pinnacle of ace-trades. Teams will forever struggle to find a return as significant as Lee, Phillips and Sizemore, three players at premium positions who didn?t just work out, but turned into superstars.

Cleveland ended up selling Phillips low, and he went on to become the second-greatest second baseman in Reds history.

They got a Cy Young season from Lee in 2008 but dealt him to the Phillies for very little in 2009.

Sizemore was on his way to becoming a perennial All-Star, but injuries have limited him to 104 games the last four seasons.

It?s almost unthinkable that Montreal traded so much to get Colon, but again it?s an example of a team in win-now mode talking itself into mortgaging the future for the here and now. There will be teams that talk themselves into the very same thing this July. It?s Ruben Amaro Jr.?s job to push for the best possible offer and stand his ground if he doesn?t get at least one impact bat close to major-league ready.

Dark 'Man of Steel' groomed to reboot Superman film franchise

LOS ANGELES (Reuters) - "Man of Steel," the latest iteration of the Superman series, lands on the big screen on Friday with the hopes that the dark, action-heavy film can revive the classic comic book superhero as a Hollywood franchise.

But director Zack Snyder, who is best known for 2007 action film "300" and 2009 superhero flick "Watchmen," told Reuters that it was a delicate balance staying faithful to Superman's story without retreading earlier film and television versions.

"I think a mistake would be to consciously ask, 'What mistakes have they done in the past?' That type of reactive approach - that's going to end in disaster," Snyder, 47, said.

"The way we looked at it was like, 'Let's say we just found these comic books under our bed and this character, Superman, seems pretty cool to make a movie about,'" he added.

The Superman comic, which artist Joe Shuster and writer Jerry Siegel debuted in 1938, has been brought to the big and small screen about a dozen times, most notably in the four-film series starring Christopher Reeve from 1978 to 1987.

"Man of Steel" tells the origin story of the DC Comics superhero, played by British actor Henry Cavill, setting up the film as a springboard to reboot a franchise that failed to take off in 2006 with director Bryan Singer's "Superman Returns.

Studio Warner Bros. enlisted director Christopher Nolan - whose three Batman films grossed more than $2 billion worldwide between 2005 and 2012, according to box-office tracker Boxofficemojo.com - as a producer and story writer to help shepherd the film along.

"He treated me probably how he would want to be treated," Snyder said of Nolan. "If I had a problem he was there to say, 'What's going on? What about this?' offering advice and counsel whenever I needed it."

ALL-AMERICAN ALIEN

"Man of Steel" begins with Superman's birth as Kal-El on the planet Krypton. As the last hope for his society's survival amid an insurrection, his parents send him to Earth where he is raised as Clark Kent in Smallville, Kansas.

By framing "Man of Steel" as a coming-of-age story, Snyder is also able to tap the science-fiction unique to Superman that other superhero stories, like Batman, do not have.

"He's able to look at humanity objectively because he's not human, and that element combined with the sci-fi school made me really go after that aspect of it as hard as I could," he sal elaid.

The film documents the young life of Clark Kent from bullied and sensitive schoolboy to itinerant loner endowed with otherworldly strength, laying the groundwork for a consistent, relatable Superman that Snyder said he wanted in the character.

"We didn't have to have this crazy transformation go on when he became Clark Kent," he said. "He's not even aware of it ... I feel like in a weird way, he's also easier to hook onto because he's coming from the same point of view all the time."

Snyder also said he leaned on Clark Kent's all-American values and farm-boy upbringing by Jonathan (Kevin Costner) and Martha Kent (Diane Lane) when it came time for Superman to emerge to save the planet from Kryptonian warlord General Zod.

"He's like a first-responder. ... He's a volunteer in a lot of ways, and I think that because he's that ultimate, selfless character, you end up (with) a personality type that goes along with that," Snyder said.

"I think as soon as you start to work on him it's going to be muddy and weird," Snyder said about his reluctance to modify Superman from his comic book origins. "There's a pure thing inside of the Superman character."

"Man of Steel" also stars Amy Adams as Lois Lane and Michael Shannon as General Zod.
